平菇子实体钙调素的分离纯化及其与猪脑钙调素的生物活性比较

平菇萃取液经酸性沉淀、热变性、Phenyl-Sepharose CL-4B疏水亲和层析和DEAE-Cellulose 52离子交換层析分冉出了电泳纯的真菌钙调素。在比较钙调素对磷酸二酯酶活化的能力和ELISA实验的免疫亲和力对发现,平菇钙调素与猪脑钙调素的生物活性有较大差异,提示在钙调素定量测定中有必要考虑到标准钙调素与样品钙调素之间的同源性差异。     

植物凝集素(PHA)对小鼠白细胞及血清乳酸脱氢酶同工酶的影响

本文就 PHA 在不同时间内对小鼠白细胞及血清 LDH,同工酶的影响进行了观察,结果表明,PHA 对小鼠白细胞及血清的 LDH 酶活性有一定的影响,24h,48h,72h 三个不同时间组均与对照组有显著性差异(P<0.01),其中以48h 组酶活性最强,同时淋巴细胞的酶活性明显高于粒细胞;血清同工酶谱也有明显的变化,LDH_(1-4)均低于正常(P<0.01),LDH_5明显增高(P<0.01),同时出现了 LDH_5(?)亚带.     

Diverse flowering responses subjecting to ambient high temperature in soybean under short-day conditions

Flowering time is one of important agronomic traits determining the crop yield and affected by high temperature. When facing high ambient temperature, plants often initiate early flowering as an adaptive strategy to escape the stress and ensure successful reproduction. However, here we find opposing ways in the short-day crop soybean to respond to different levels of high temperatures, in which flowering accelerates when temperature changes from 25 to 30 °C, but delays when temperature reaches 35 °C under short day. phyA-E1, possibly photoperiodic pathway, is crucial for 35 °C-mediated late flowering, however, does not contribute to promoting flowering at 30 °C. 30 °C-induced up-regulation of FT2a and FT5a leads to early flowering, independent of E1. Therefore, distinct responsive mechanisms are adopted by soybean when facing different levels of high temperatures for successful flowering and reproduction.     

Thioredoxin 1 overexpression attenuated diabetes-induced endoplasmic reticulum stress in Müller cells via apoptosis signal-regulating kinase 1

As one of the common and serious chronic complications of diabetes mellitus (DM), the related mechanism of diabetic retinopathy (DR) has not been fully understood. Müller cell reactive gliosis is one of the early pathophysiological features of DR. Therefore, exploring the manner to reduce diabetes-induced Müller cell damage is essential to delay DR. Thioredoxin 1 (Trx1), one of the ubiquitous redox enzymes, plays a vital role in redox homeostasis via protein–protein interactions, including apoptosis signal-regulating kinase 1 (ASK1). Previous studies have shown that upregulation of Trx by some drugs can attenuate endoplasmic reticulum stress (ERS) in DR, but the related mechanism was unclear. In this study, we used DM mouse and high glucose (HG)-cultured human Müller cells as models to clarify the effect of Trx1 on ERS and the underlying mechanism. The data showed that the diabetes-induced Müller cell damage was increased significantly. Moreover, the expression of ERS and reactive gliosis was also upregulated in diabetes in vivo and in vitro. However, it was reversed after Trx1 overexpression. Besides, ERS-related protein expression, reactive gliosis, and apoptosis were decreased after transfection with ASK1 small-interfering RNA in stable Trx1 overexpression Müller cells after HG treatment. Taken together, Trx1 could protect Müller cells from diabetes-induced damage, and the underlying mechanism was related to inhibited ERS via ASK1.     

别藻蓝蛋白藻蓝胆素发色团分子构象研究

主要研究了蓝绿藻污棕席藻(Phormidium luridum)别藻蓝蛋白在不同 pH值条件下的吸收光谱和共振拉曼光谱.发现低聚化的结果导致了三聚体别藻蓝蛋白 650nm 特征吸收峰的消失和一些共振拉曼带强度和位置的移动.结果表明在低 pH 值作用下的低聚化的别藻蓝蛋白中藻蓝胆素发色团分子的构象和自由胆素分子类似,比三聚体的别藻蓝蛋白的发色团分子更趋于卷曲,折叠的构象态.而三聚体的别藻蓝蛋白,主要的拉曼带 1645cm^-1是其发色团分子构象处于更线性延展的标志,其光谱行为和吸收光谱 A_vis/A_uv所表征的发色团分子构象的结果相一致.     

侧脑室注射促甲状腺激素释放激素对大鼠睡眠—觉醒的影响

采用多导睡眠描记术研究了例脑室注射促甲状腺激素释放激素(TRH)对正常大鼠和去甲状腺大鼠睡眠-觉醒的影响。在正常大鼠,TRH引起觉醒增加,浅慢波睡眠(SWS_1)、深慢波睡眠(SWS_2)和总睡眠时间(TST)均减少,异相睡眠(PS)消失,SWS_1、SWS_2和PS的潜伏期均显著延长,给药后立即产生效应并在1h内达高峰。去甲状腺对大鼠的睡眠-觉醒无明显影响,注射TRH后引起的效应与正常大鼠相似。结果提示TRH有促进大鼠觉醒的作用,对各睡眠时相均有抑制作用,其作用部位可能在下丘脑以外的中枢结构。     

Solid-phase microextraction and cuticular hydrocarbon differences related to reproductive activity in juniper bark borer Semanotus bifasciatus Motschulsky

Cuticular hydrocarbons of Cerambycidae species can function as signals for sex recognition. Little is known about the copulatory signals of the juniper bark borer Semanotus bifasciatus, a major economic threat to Platycladus orientalis Franco in China. Here, we investigated the cuticular hydrocarbons of both sexes of S. bifasciatus to determine the chemically mediated mating signals using the solid-phase microextraction (SPME) technique with carbowax/divinylbenzene fibers (CAR/DVB) and then analyzed by coupled gas chromatography-mass spectrometry (GC-MS). A series of aliphatic saturated straight-chain n-alkanes (n-C₂₃ to n-C₂₈), internally branched monomethylalkanes at carbons 3, 11, or 13, and dimethylalkanes were identified, which showed no qualitative differences in either sex and were similar in the samples with SPME fiber extraction and those with hexane extraction. The bioassay showed that 11-methylpentacosane (11-MeC₂₅), 11-methylhexacosane (11-MeC₂₆), and 11-methylheptacosane (11-MeC₂₇) have sex-specific recognition functions that triggered more mating attempts at a female-specific ratio of 100:4:60 than at a male-specific ratio of 100:85:50. In addition, the female-specific ratio of 11-methylalkanes can elicit about 70% of male mating attempts within about 60 s, whereas live females elicit about 98% of male mating attempts within 25 s. The discrepancy in the initiation of mating attempts by synthetic mixtures and live females suggests that the methyl isomers 3-MeC₂₅, 3-MeC₂₇, and/or 11,15-diMeC₂₇ may also be involved in the mating behavior of S. bifasciatus. These results suggest that 11-MeC₂₅, 11-MeC₂₆, and 11-MeC₂₇ constitute the contact sex pheromone of S. bifasciatus, with the presence or absence of 11-MeC₂₆ in particular playing an important role in mate recognition by males.
